Intro prompt need to be recorded without music, PBX mix the MOH and intro prompt , this way your audio is good

If intro prompt is already recorded with music and client doesn't want to pay for another one (wich is perfectly understandable), we set the intro prompt as MOH in this queue (only in this queue, not in general settings), and let intro prompt empty.
This way the client hear intro prompt correctly, only downside is its played again as long as the caller is in the queue but its not really an issue as you can prevent this by switching to another queue after the message as being played, second queue with same settings except different moh.

The intended way of operation by design is for the intro prompt to start playing with the MOH simultaneously.

The MOH volume is lower at that moment, and when the intro is over the volume goes back to normal, so that they are mixed together for a nicer effect.

If you want to play a separate message, you can send the caller to an IVR, call it say "Intro Message" and then make the IVR send the call to your queue.

Another option would be to use a playlist as MOH. In the playlist, the first music file can be silence for a few seconds (to cover for the intro prompt), and the second file can be a music file that is a bit longer than the Maximum Queue Wait Time. The customer will never hear it loop in this case

Sorry JohnS but if you use an IVR, the caller will always hear all the intro prompt message before it start ringing wich is very annoying and clients dont want this most of the time.
I think if the intro prompt has music on it, the best way is to do as I said before, put this intro prompt in queue moh and nothing in queue intro prompt. See my post before.
 
I think Khaled was going for something different, he does want it to play everytime someone call that queue but just once and separately from the MOH before the music starts
