I've experience the same "no audio issue" mentioned here:
https://www.3cx.com/community/threads/windows-client-issue-no-audio-microphone-works.76666/
However, in my case the problem is solved because removing the Realtek audio drivers was only a temporary fix. The problem is: all different USB headsets work fine in any application including the 3CX webclient(!) but in 3CX Softphone for Windows the microphone and speakers seem to be completely muted as soon as you start a call. We tested this with the echo service (*777) as well as actual calls. Apparently, 3CX Softphone for Windows is doing something different than the other applications using the audio device. My guess is that Windows or some driver mutes the device when it detects "communication". However this Windows sound setting is already disabled.
What's even worse: When this issue occured on an older notebook, I've set up another one - completely different vendor and model. I made a short test with 3CX Softphone and a headset and everything was fine. Also, 3CX Softphone had been used before on this notebook until just two days ago. But when I was finished with updating all software, the same issue appeared on this notebook as well. As a workaround, I've set up the Chrome 3CX extension for the user, which is fine in this case.
Nonetheless, I'm worried that some morning none of my colleagues can use their softphones as turned all of them silent due to a (Windows?) update.
The affected notebooks:
Thinkpad Edge 13
Fujitsu Lifebook S751
App: 3CXPhone for Windows 16.3.179
OS: Windows 10 20H2 (Build: 19042.630) but the issue was already in Windows 10 19xx.
