What is and what can be reason to enable keep alive function in settings > network > firewall ?

Its normally used to keep NAT port mappings alive. Not really required if you've set up your static mappings correectly.

enable applications using the Real-time Transport Protocol (RTP) and the RTP Control Protocol (RTCP) to keep their RTP Network Address Translator (NAT) mappings alive.
 
Specifically it will have your PBX send a few UDP packets from its SIP Port to the SIP providers every so many seconds. As @kieferschild mentioned though, it should not be required if the firewall is configured correctly.
