I have been reading through the documentation but I don't see this as a feature.

Is it possible to set DND status via a 3CXPhone HTTP API?

No this is not possible through HTTP API, only what is listed in the ref. page is.
On the other hand it might be possible with Call Control API, have a look at the .chm documentation for v12 on http://www.3cx.com/blog/docs/call-control-api/
